Summary

On 11 March 2020, the World Health Organization declared COVID-19 a global pandemic, and the disease now affects nearly every country and region. Caused by SARS-CoV-2, COVID-19 continues nearly 18 months later to present significant challenges to health systems and public health in both hemispheres, as well as the economies of every country. The morbidity and mortality of the infection caused by SARS-CoV-2 has been significant, and various waves of disease outbreaks initially overwhelmed many hospitals and clinics and continue to do so in many countries. This influences everyone, and public health countermeasures have been dramatic in terms of their impact on employment, social systems, and mental health. This entry collection aims to gather diverse fields about COVID-19, including in epidemiology, public health, medicine, genetics, systems biology, informatics, data science, engineering, sociology, anthropology, nursing, environmental studies, statistics, and psychology.

COVID-19 and Digital Transformation
COVID-19 is a driver of digital transformation. Technology, automation, and collaboration (TAC) is the most positive significant factor which enables work from anywhere (WFA) (or work from home) arrangements and also leads to the third positive factor of a work-life balance (WLB). The top three negative factors are no work-life balance (NWL), social employment issues (SEI), and data security and technology issues (DST). While the pandemic situation is leading to a positive situation for economies and organizations at a micro level, the negative impacts, which will affect overall economic growth as well as social, health, and wealth wellbeing, need to be kept in mind.

SARS-CoV-2 and Acute Cerebrovascular Events
Since the coronavirus disease 2019 (COVID-19) pandemic, due to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, accumulating evidence indicates that SARS-CoV-2 infection may be associated with various neurological manifestations, including acute cerebrovascular events (i.e., stroke and cerebral venous thrombosis). These events can occur prior to, during and even after the onset of COVID-19's general symptoms. Although the mechanisms underlying the cerebrovascular complications in patients with COVID-19 are yet to be fully elucidated, the hypercoagulability state, inflammation and altered angiotensin-converting enzyme 2 (ACE-2) signaling in association with SARS-CoV-2 may play key roles. ACE-2 plays a critical role in preserving heart and brain homeostasis. As the number of published COVID-19 cases with cerebrovascular events is growing, prospective studies would help gather more valuable insights into the pathophysiology of cerebrovascular events, effective therapies, and the factors predicting poor functional outcomes related to such events in COVID-19 patients.

Inclusive Leisure during COVID-19 Pandemic
La pandemia de COVID-19 ha provocado una reducción de las actividades de ocio que implican contacto humano. El aislamiento social se ha incrementado, especialmente entre las personas vulnerables con una red de apoyo frágil, como es el caso de los jóvenes que han abandonado el cuidado. El objetivo de la presente investigación fue identificar propuestas e intervenciones socioeducativas implementadas durante la pandemia relacionadas con el ocio como forma de promover la inclusión social de estos jóvenes. Para ello, se realizó un estudio cualitativo en el que se realizaron veinte entrevistas semiestructuradas a jóvenes que habían abandonado el sistema de atención, además de quince entrevistas a profesionales que trabajan con este colectivo en la realización de intervenciones socioeducativas. El análisis del discurso reveló que el aislamiento por la crisis de salud tuvo mayores repercusiones en entornos normalizados en los que la actividad de ocio se redujo con este creciente riesgo de inclusión social entre estos jóvenes. Las propuestas y experiencias que surgen de este entorno evidencian que las intervenciones socioeducativas dirigidas al ocio facilitan la inclusión social. En este sentido, se sugieren futuras líneas de investigación para optimizar los resultados de las intervenciones socioeducativas dentro de este grupo.

The second year of the COVID-19 (coronavirus disease) pandemic has seen the need to identify and assess the long-term consequences of a SARS-CoV-2 infection on an individual’s overall wellbeing, including adequate cognitive functioning. ‘Cognitive COVID’ is an informal term coined to interchangeably refer to acute changes in cognition during COVID-19 and/or cognitive sequelae with various deficits following the infection. These may manifest as altered levels of consciousness, encephalopathy-like symptoms, delirium, and loss of various memory domains. Dysexecutive syndrome is a peculiar manifestation of ‘Cognitive COVID’ as well.

Age-Friendly Built Environment
Age-friendly built environments have been promoted by the World Health Organisation (WHO, Geneva, Switzerland) under the Global Age-friendly Cities (AFC) movement in which three domains are related to the built environment. These are: housing, transportation, outdoor spaces and public buildings. The aim is to foster active ageing by optimising opportunities for older adults to maximise their independent living ability and participate in their communities to enhance their quality of life and wellbeing. An age-friendly built environment is inclusive, accessible, respects individual needs and addresses the wide range of capacities across the course of life. Age-friendly housing promotes ageing in familiar surroundings and maintains social connections at the neighbourhood and community levels. Both age-friendly housing and buildings provide barrier-free provisions to minimise the needs for subsequent adaptations. Age-friendly public and outdoor spaces encourage older adults to spend time outside and engage with others against isolation and loneliness. Age-friendly public transport enables older adults to get around and enhances their mobility. For achieving an age-friendly living environment, a holistic approach is required to enable independent living, inclusion and active participation of older adults in society. The eight domains of the AFC movement are not mutually exclusive but overlap and support with one another.

Nanotechnology-Enabled COVID-19 mRNA Vaccines
COVID-19 mRNA vaccines contain synthetic mRNA sequences encoded for the Spike proteins expressed on the surface of SARS-CoV-2, and utilize the host cells to produce specific antigens that stimulate both humoral and cellular immunities. Lipid nanoparticles are essential to facilitate the intracellular delivery of the mRNA to its action site, the ribosome, to fully exert its effect.

Polydatin/Resveratrol interference in ACE2:Spike recognition
In the search for new therapeutic strategies to contrast SARS-CoV-2, we here studied the interaction of polydatin (PD) and resveratrol (RESV)—two natural stilbene polyphenols with manifold, well known biological activities—with Spike, the viral protein essential for virus entry into host cells, and ACE2, the angiotensin-converting enzyme present on the surface of multiple cell types (including respiratory epithelial cells) which is the main host receptor for Spike binding. Molecular Docking simulations evidenced that both compounds can bind Spike, ACE2 and the ACE2:Spike complex with good affinity, although the interaction of PD appears stronger than that of RESV on all the investigated targets. Preliminary biochemical assays revealed a significant inhibitory activity of the ACE2:Spike recognition with a dose-response effect only in the case of PD.

SARS-CoV-2 Detection from Surface Samples
Contaminated surfaces have been discussed as a possible source of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). Under experimental conditions, SARS-CoV-2 can remain infectious on surfaces for several days. However, the frequency of SARS-CoV-2 detection on surfaces in healthcare settings and the public is currently not known. A systematic literature review was performed. On surfaces around COVID-19 cases in healthcare settings (42 studies), the SARS-CoV-2 RNA detection rates mostly were between 0% and 27% (Ct values mostly > 30). Detection of infectious SARS-CoV-2 was only successful in one of seven studies in 9.2% of 76 samples. Most of the positive samples were obtained next to a patient with frequent sputum spitting during sampling. Eight studies were found with data from public surfaces and RNA detection rates between 0% and 22.1% (Ct values mostly >30). Detection of infectious virus was not attempted. Similar results were found in samples from surfaces around confirmed COVID-19 cases in non-healthcare settings (7 studies) and from personal protective equipment (10 studies). Therefore, it seems plausible to assume that inanimate surfaces are not a relevant source for transmission of SARS-CoV-2. In public settings, the associated risks of regular surface disinfection probably outweigh the expectable health benefits.
